Methamphetamin ist ein potenteres Analogon von Amphetamin mit höherer Lipophilie und schnellerer ZNS-Penetration. Der Wirkmechanismus ist analog zu Amphetamin: TAAR1-Agonismus → PKA/PKC-Aktivierung → Phosphorylierung von DAT und NET → massiver Dopamin- und Noradrenalin-Reverse-Transport. Zusätzlich VMAT2-Inhibition setzt vesikuläre Monoamine frei. Im Vergleich zu Amphetamin: stärkere Dopaminfreisetzung, längere Halbwertszeit (~10-12h vs. 8-12h), höheres Abhängigkeitspotential und stärkere neurotoxische Effekte. Die Methyl-Gruppe erhöht die Lipophilie und führt zu schnellerem Wirkungseintritt und intensiveren Effekten.
