Additive μ-Opioid-Wirkung und Atemdepression
Tianeptin wirkt als atypisches Antidepressivum mit μ-Opioid-Rezeptor-Agonismus und AMPA-Rezeptor-Modulation. Die opioidartige Pharmakologie führt in Kombination mit anderen Opioiden zu einer additiven Wirkung mit erhöhtem Risiko für Atemdepression. Tianeptin wird zunehmend missbräuchlich in hohen Dosen konsumiert.
Additive ZNS-Depression durch Opioid-GABAerge Potenzierung
Die μ-Opioid-Wirkung von Tianeptin in Kombination mit der GABAergen Potenzierung durch Benzodiazepine führt zum gleichen Mechanismus wie bei klassischen Opioid-Benzodiazepin-Kombinationen: synergistische Atemdepression.
